Checking the fold: fast tests that show a design folds as drawn

Abstract

A design is useful only if it folds into the shape the model drew. We describe a one-day screen that uses circular dichroism (CD) and size-exclusion chromatography (SEC) on 384 designs at a time. In a test set of 1,152 designs, 91% had the expected secondary structure and a single main peak.
